Many and plenty depending on the drug. Painkillers can cause liver problems. They alone, or used in combination with sedatives, can depress your central nervous system and cause respiratory failure. It is far more common than most people think. Kidneys are at risk too, as is the stomach. They can cause esophageal spasms that make eating and swallowing difficult. Most will lead to severe depression that takes a long time to get through. Not to mention the horrible physical withdrawal symptoms and the long mental recovery and cravings. Oh, cardiac arrest is possible too, painkillers or stimulants can cause this.
This is an incomplete list and just what I can think of of the top of my head. But just this includes liver, kidneys, stomach and digestive system, the nervous system, the brain (depression) and the heart.
